Dr. Frederick Frankenstein (Gene Wilder) is a respected professor at a medical school, and is engaged to a woman named Elizabeth (Madeline Kahn). He always insists, when anyone brings up his late grandfather, that his name be pronounced "Fronk'-en-steen". Frankenstein soon learns that he has inherited the estate of his family in Transylvania, and travels there to claim his property. Upon arrival he meets his new lab assistant Inga (Teri Garr), and the estate's servants Frau BlГјcher (Cloris Leachman) and Igor (Marty Feldman). Later, Dr. Frankenstein reads his grandfather's journals and is inspired to continue his experiments in reanimating the dead.

After exhuming the body of a dead criminal, he puts Igor to the task of obtaining the brain of a famous dead scientist. But things don't go as planned, and instead he returns with a brain labeled "Do Not Use This Brain! Abnormal". Dr. Frankenstein is successful in breathing life into his creation of body and mind, but his experiment quickly gets out of control with his new creature experiencing the world in innocent yet destructive ways.
